The dopaminergic neuronal cell line (SN) from transgenic mouse embryos was the first cell culture model to be tested for gene expression. These cells maintain many essential features of dopaminergic neuronal phenotype and cellular context. Glycosphingolipids are enriched in the cellular plasma membrane and are thought to play a vital role in normal mammalian embryogenesis. The major objective of the proposed research is to characterize the effects of ethanol on glycosphingolipid metabolism in gastrulation and organogenesis stage mouse embryos grown in culture.

Embryonic stem cells (ESCs) are found in the inner cell mass of the human blastocyst, an early stage of the developing embryo lasting from the 4 th to 7 th day after fertilization. In normal embryonic development, they disappear after the 7 th day, and begin to form the three embryonic tissue layers. ESCs extracted from the inner cell mass during the blastocyst stage, however, can be cultured. Nov 08,  · The inner cell mass of pre-implantation blastocyst stage embryos is a source of cells that can be cultured indefinitely in vitro as a self-renewing, pluripotent population. In this review, we Cited by:

Differentiated cells can be reprogrammed to an embryonic-like state by transfer of nuclear contents into oocytes or by fusion with embryonic stem (ES) cells. Little is known about factors that induce this reprogramming. Here, we demonstrate induction of pluripotent stem cells from mouse embryonic or adult fibroblasts by introducing four factors, Oct3/4, Sox2, c-Myc, and Klf4, under ES cell Cited by: A variety of glycoconjugates, including glycosphingolipids (GSLs), expressed in mammalian tissues and cells were isolated and characterized in early biochemical studies.

That plasma membrane preparations of various cultured animal cells are significantly enriched in these compounds () and also from immunological studies (25). Partial characterization of the gangliosides of monkey kidney cells (22), mouse L-cells (23, 26), mouse embryo secondary cultures (26), various mouse.

In recent years the knowledge regarding glycosphingolipids in human embryonic stem cells has been extended by biochemical studies, which is the focus of this review.

In addition, the distribution of the human pluripotent stem cell glycosphingolipids in human tissues. Jun 21,  · Glycosphingolipids were also characterized by MALDI-TOF/TOF MS of oligosaccharides released from glycosphingolipids by enzyme treatment in a highly ambitious study of the total cellular glycome (glycosaminoglycans, N- and O-linked saccharides, and glycosphingolipids) of human pluripotent stem cell lines (hESC and hiPSC).

However, in this study the cells were cultured on Cited by: 4. Glycosphingolipids (GSLs) are a type of glycolipid. They are found in the cell membranes of organisms from bacteria to man, and are the major glycans of the vertebrate brain, where more than 80% of glycoconjugates are in the form of glycolipids.

The emphasis of this chapter is on vertebrate glycosphingolipids. In general, the hexosamine-containing macromolecules of eukaryotic cells may be classified as glycosphingolipids, glycoproteins, and glycosaminoglycans.

Since there is no available information concerning the synthesis of glycoproteins by clonal lines derived from the nervous system in tissue culture, the metabolism of this class of substances will not be considered in this dentalimplantsverobeach.com by: Dramatic changes in glycolipid synthesis from one series to another series have been observed to be associated with early mouse embryogenesis and with differentiation of mouse leukemia cells.

Finally, some glycosphingolipids occur on the cytosolic surface of cellular membranes.

Amazingly, their fate and function is unknown which illustrates that we solved only part of the puzzle posed by these sphinx-like Cited by: Dec 31,  · In this study, Ugcg expression was reduced to approximately 80% by short hairpin RNAs (shRNAs) to evaluate the roles of glycosphingolipids in proliferation and neural differentiation of mouse embryonic stem cells (mESCs).Cited by: Two lines of studies clearly established the occurrence of CCI mediating adhesion of mouse embryonal cells, by us [87,], and CCI mediating species-specific autoaggregation of sponge cells, by Max Burger's group [–].Cited by: GD3 is a predominant ganglioside in embryonic brains and in primary neural precursor cells (9, 25), but the neural precursor cells prepared from ST-II-knockout mice exhibit little or no phenotype.

These data strongly suggest that the functions of b-series gangliosides may be compensated for by the remaining GSLs, such as a-series dentalimplantsverobeach.com by: NGF has a variety of effects on the central nervous system (CNS) under experimental conditions: It enhances the regeneration of central adrenergic neurons and it brings forth temporal changes in surface specificity of cultured embryonic chick tectal cells.

Apr 05,  · Isolation of Glycosphingolipids from Human Embryonic Stem Cell Lines. Non-acid glycosphingolipids were isolated from the two hESC lines (SA and SA), essentially as described (12, 13). The cells (1 × 10 9 cells from each cell line) were extracted by the addition of first 5 ml of methanol at 70 °dentalimplantsverobeach.com by: Jun 12,  · The easy generation and physiological relevance of mouse embryonic fibroblasts (MEFs) have made them a powerful tool in discovering novel signaling pathways, investigating regulatory networks, and exploring biochemical profiling of protein complexes involved in Cited by: 8.

Mouse embryonic fibroblasts (MEFs) are the most commonly used feeder cell type and have reliably served as feeder cells for co-culture with mouse ESC since they were first derived in This chapter provides examples of alternatives to culture of hESC on MEF feeder layers.

The physiological role of GSLs has been studied using genetic, biochemical, biophysical and cell biology approaches. Mouse genetics has provided a general framework for our understanding of the roles of GSLs in mammals. According to these studies, ablation of the gene for GlcCer synthase in mice leads to embryonic lethality during gastrulation Cited by: Induction of a Deoxycytidineless State in Cultured Mammalian Cells by Bromodeoxyuridine Expression of Thymidine Kinase Variants is a Function of the Replicative State of Cells Book Reviews Among the Protozoologists Cell Culture-The State of the Art Ribonucleic Acids in Review J.

Until now, there has never been such a comprehensive compendium, though there have been more focused books of protocol, such as Manipulating the Mouse Embryo, from Cold Spring Harbor. This book is intended to appeal to all constituencies, from basic.

Glycosphingolipids are thought to provide protection from harsh conditions in the extracellular environment, such as low pH and degrading enzymes. They help cells to interact with extracellular matrices and other cells, and serve as surface receptors for bacterial toxins and possibly endogenous extracellular molecules.
